Need wisdom of biochem grad students:

Is it possible to hypermethylate promoters and/or enhancers in vivo to induce gene silencing in a lab? Further, methylating the promoter would usually(?) confer greater silencing over the enhancer because it would directly impede basal levels of transcription?

I need to argue that methylating a promoter vs an enhancer will affect transcription levels differently enough to distinguish between the two elements. Is this there a case like this that has been tested? I can not find one for the life of me.
